SPlink Detection Kit (ABC for IHC)
Streptavidin is a protein that has similar binding properties to avidin.It is isolated from streptomyces avidinii. Streptavidin has a molecular weight of 60 KD and has 4 subunits. Each subunit can bind one molecule of biotin.SPlink Detection Kit is coupled steptavidin with horseradish peroxidase (HRP). This highly sensitive detector conjugate binds to biotinylated secondary antibodies in immunohistochemistry.

IHC Detection Procedure

Section paraffin,dewax conventionally

Antigen retrieval (this step is optional according to the experiment requirement)

Incubate with 3% H2O2 deionized water for 5~10min to eliminate endogenous peroxidase activity.

Wash with PBS, 3 min, 3 times

Add reagent A, incubate at room temperature for 10~15min, abandon the reagent, don’t wash;

Add primary antibody with proper dilution ratio,incubate at 37℃ for 2~3h or at 4℃ for one night

Wash with PBS, 3 min, 3 times

Add reagent B, incubate at 37℃ or room temperature for 10~15min

Wash with PBS, 3 min, 3 times

Add reagent C, incubate at 37℃ or room temperature for 10~15min

Wash with PBS, 3 min, 3 times

Perform chromogenic reaction with chromogenic reagent (DAB or ACE)

Wash with tap water
-
Make the section counterstained,dewatered, transparent if the experiment requires

Mount the section with proper mounting medium
Storage
Stored at 2°C – 8°C. Shading light.
