Ribonuclease H

Ribonuclease H (RNase H) specilically degrades the RNA strand from RNA:DNA hybrids. It does not hydrolyse the phosphodiester
bond from single-stranded or double stranded DNA or RNA. RNase H is purified from E.Coli expressing the recombinant rnhA gene. The recombinant protein has a molecular mass of 17 kDa and can be heat-inactivated by incubation at 65°C for 20 min.

Applications for RNase H

• Removal of RNA after first-strand synthesis by reverse transcription to allow for second strand synthesis reaction.
• Identification of RNA:DNA hybrids
