Neurofilaments can be defined as the intermediate or 10nm diameter filaments found in neuronal cells. They are composed as a mixture of subunits which often includes the neurofilament triplet proteins, NF-L, NF-M and NF-H. Neurofilaments may also include peripherin, α-internexin, nestin and in some cases vimentin. Antibodies to the various neurofilament subunits are very useful cell type markers since the proteins are quite abundant, biochemically stable. To raise this antibody bovine intermediate filaments were prepared from spinal cords by the method of Delacourte et al. and the cytoskeletal material was dissolved in 6M urea. Individual neurofilament subunits were purified by ion exchange chromatography on DEAE cellulose followed by preparative gel electrophoresis. The form of NF-H isolated is the heavily phosphorylated axonal variant, and the antibody generated recognizes predominantly axonal neurofilaments.
